On Sunday (January 5), the 62-year-old actress appeared at the prestigious ceremony which took place at the Beverly Hilton in Beverly Hills, California.
Moore was one of the shining stars at the show, winning the award for Best Performance by an Actress in a Motion Picture – Musical or Comedy, for her critically acclaimed performance in body horror The Substance, which was released last year.
Her victory came against a tough field of contenders, including Cynthia Erivo for Wicked, Mikey Madison for Anora, and Karla Sofía Gascón for Emilia Perez.
The actress, known for her iconic roles in films like Ghost, Indecent Proposal, and A Few Good Men, expressed genuine surprise at her win and proceeded to deliver a powerful speech about her 45-year acting career.
Watch the moment below:In her emotional speech, Moore shared that she had been in the industry for over four decades, and yet this "is the first time" that she's ever won "anything as an actor".
She then opened up about the fact she considered quitting the film business before being cast in The Substance - which follows an aging celebrity who uses a black-market drug to create a much younger version of herself after being fired by her producer.
“Thirty years ago, I had a producer tell me I was a popcorn actress, and, at that time, I made that mean this wasn’t something I was allowed to have," she revealed. "I could do movies that were successful, that made a lot of money, but I couldn't be acknowledged and I bought in and I believed that.
"That corroded me over time to the point where I thought a few years ago that maybe this was it. Maybe I was complete, maybe I'd done what I was supposed to do," she continued. "And when I was at kind of a low point, I had this magical, bold, courageous, out-of-the-box absolutely bonkers script come across my desk called The Substance, and the universe told me you’re not done."

The mom-of-three expressed gratitude to the movie's director Coralie Fargeat, her co-star Margaret Qualley, and “the people who’ve been for me for over 30 years... all the people who stood by me, especially the people who believed in me when I haven’t believed in myself".
“I’ll just leave you with one thing I think this movie is imparting,” she said. “In those moments when we don’t think we’re smart enough or pretty enough or skinny enough or successful enough or basically just not enough, I had a woman say to me, ‘Just know, you will never be enough, but you can know the value of your worth if you just put down the measuring stick.’
“And so today I celebrate this as a marker of my wholeness, and of the love that is driving me, and the gift that is doing something I love and being reminded that I do belong. Thank you so much,” she concluded.
The actress's incredible message received extended applause from the crowd, and viewers watching at home.
"Such a beautiful speech. So happy to see her get her well-deserved nod," one fan wrote, while a second shared: "It baffles me to think that this is her first award. Not only am I happy to see her finally getting her flowers, but to see her win for this role specifically? Beyond deserved."
"One of the best speeches I’ve heard in a long time. She deserves an Oscar, too," a third added.
According to the BBC, Moore's Golden Globe win may increase her chances of an Oscar nomination.
